Cancer is a complex disease involving various genes and cellular processes. Therefore, to eliminate cancer cells, a combination of treatments with different targets is required. In this review, a new potential therapeutic approach is addressed: inhibition of a single RNA degradation pathway to inhibit tumorigenesis by acting on different cellular processes and the expression of different genes. Re-expression of different genes is also discussed: tumor suppressor genes with a nonsense mutation, oncogenes suppressed by NMD, and genes that induce a specific anti-cancer immune response. Finally, putative limitations of cancer treatment by NMD inhibition are addressed.
